The track, performed by Korean American artists Ejae, Audrey Nuna and Rei Ami, was announced as the winner at the ceremony held at the MGM Grand Garden Arena in Las Vegas on Monday (U.S. time).
It beat nine other nominees, including Taylor Swift's "The Fate of Ophelia," to take the top honor.
"Truly, like this movie, this song is fueled by the fans. You guys filled the Honmoon," Ejae said in her acceptance speech on stage, expressing appreciation to fans and the "KPop Demon Hunters" team. "Thank you to Rei," she added. "Happy birthday."
Rei said, "Thank you to our friends, family and teams and our incredible 'KPop Demon Hunters' family for a life-changing year."
The other member of the trio, Audrey Nuna, did not attend the ceremony.
Co-written by Ejae, "Golden" topped the Billboard Hot 100 for eight non-consecutive weeks and led the British Official Singles Chart Top 100 for 10 nonconsecutive weeks. The soundtrack album reached No. 1 on the Billboard 200 chart.
The song earlier won at major global award ceremonies, including the Grammy Awards, the Academy Awards and the Golden Globe Awards.
