"Both occupants of the vehicle, who are yet to be formally identified, died at the scene," a Victoria Police spokesperson said.
Footage from the scene shows a crumpled silver coloured car on the side of the road near the intersection of the highway and James Street.
Firefighters and the State Emergency Service were called.
The Princes Highway is expected to be closed for most of this morning while investigations continue.
Police warned of traffic disruptions in both directions between Robinson and Potter streets.
Detectives were still trying to establish the exact circumstances surrounding the crash.
