Officers were called shortly before 5am today to a home at Ronald Road, Taree, where they found a man who had been stabbed.
The man, who has not been formally identified, was treated by police officers and NSW Ambulance paramedics, but died at the scene.
The Manning Great Lakes police district commander, Superintendent Grant Erickson, said an investigation was underway with assistance from the State Crime Command's Homicide Squad, under Strike Force Hexalon.
He said two crime scenes had been established, at the home and at another location on Ronald Road, which were being examined by specialist forensic investigators.
"Evidence obtained from the scene would indicate there is two crime scenes," he said.
" We have teams of detectives on scene and also canvassing the surrounding areas at this stage. "
Superintendent Erickson would not confirm if the man was at home alone.
"At this stage we have witnesses who have come forward, and they are being spoken to by police at this stage, I'll just leave it at that," he said.
He said the victim was not known to police and it was unclear if it was a targeted attack.
"This investigation is very early and it would be remiss of me to speculate at this time," he said.
"At this stage, the offender is unknown."
