研究团队通过“形态学预筛+质谱法动物种属(ZooMS)鉴定”的创新高效筛选策略,最新从云南蝙蝠洞遗址6万多块碎骨中精准锁定不同地层的3件古人类化石,并联合蛋白质组测序、单氨基酸多态性分析与系统发育分析等多重分子证据,将其与4颗出土古人类牙齿中的2颗一同确认为丹尼索瓦人,年代范围为距今约16.7万年至13.4万年。

高效筛选策略
研究团队介绍,丹尼索瓦人是2010年基于遗传学分析在西伯利亚丹尼索瓦洞发现的一支古老型人类群体,广泛分布于亚洲。鉴于遗传学研究表明在大洋洲的现代人群中含有相对较高的丹尼索瓦人遗传成分,中国西南地区可能是丹尼索瓦人扩散的关键区域。
因此,在中国西南找到丹尼索瓦人化石,尤其是有鉴定特征的头后骨骼,可为解答丹尼索瓦人群的时空分布、演化历史及适应表型提供关键证据。此前,中国西南关键地域由于化石保存的原因,长期存在研究空白,制约了相关研究。
在本项研究中,基于质谱法动物种属鉴定这一大批量筛选古人类化石的首选方案,研究团队创新加入形态学预筛选流程,在6万多块碎骨中选取出22块疑似古人类化石,同时在剩余的非疑似人骨中随机挑选54个碎骨开展质谱法动物种属鉴定分析,确认在蝙蝠洞遗址进行古蛋白质分析的可行性。最终,对22块疑似人骨的质谱法动物种属鉴定分析将2块顶骨碎片和1块近端桡骨碎片确认为人属来源。
随后,研究团队进一步开展系统分析和可靠性评估,结果显示5个样本(3件骨碎片+2颗牙齿)均携带丹尼索瓦人特征性变异位点基因,支持将其可靠地归入丹尼索瓦人。其中,第四前臼齿、第二臼齿的2颗牙齿,鉴定表明均属于丹尼索瓦人男性个体。
此次研究通过“形态学预筛+质谱法动物种属鉴定”的组合策略,使古人类化石的筛选效率实现数量级跃升,也为今后在类似大规模碎骨遗址中高效发现古人类化石提供了可复制的方法。
呈现镶嵌状态
本项研究对新发现的2件丹尼索瓦人顶骨碎片进行分析显示,蝙蝠洞丹尼索瓦人顶骨的厚度与东亚中更新世晚期古老型人类、欧洲海德堡人最为接近。
作为迄今全球唯一被确认的丹尼索瓦人桡骨化石,此次近端桡骨碎片的成功鉴定,为揭示蝙蝠洞丹尼索瓦人群的颅后表型特征提供了关键证据。高精度显微CT显示,该桡骨个体拥有粗壮的骨皮质结构和完全愈合的近端骨骺,表明其属于成年或接近成年的个体。形态学分析发现,蝙蝠洞桡骨化石呈现尼安德特人与现代人的镶嵌状态,表明丹尼索瓦人在生物力学需求和行为适应模式上异于前述两个类群。
研究团队指出,丹尼索瓦人发现以来,目前通过分子证据仅鉴定到少量其他地域的丹尼索瓦人遗骸,包括中国西北青藏高原白石崖溶洞(夏河)的半个下颌骨和肋骨、中国东南台湾海峡(澎湖)的半个下颌骨,以及来自中国东北(哈尔滨)的几乎完整的头骨(龙人)。
这次研究不仅填补了丹尼索瓦人分布在中国西南的关键地理空白,还使云南蝙蝠洞成为除丹尼索瓦洞之外出土丹尼索瓦人遗骸种类最丰富的地点,且被不同时期的丹尼索瓦人使用。
丹尼索瓦人在云贵高原的生存年代虽然处于冰期,但当地相对温暖的环境和充足的食物资源,可能作为丹尼索瓦人群的狩猎场,支撑其顺利度过冰期。
研究团队表示,未来希望更多获取、整合丹尼索瓦人基因组和蛋白质组数据,进一步帮助理解丹尼索瓦人群内部形态的多样性,以及在应对气候变化和同期其他古人类竞争压力下的适应性行为,科学判断丹尼索瓦人是否或如何参与现代人的形成和扩散。
